Where to Find Twin Sesters
At Sester's Gate, go up the broken bridge and launch yourself up to the arena using the current at the end of the bridge.
Twin Sesters Attack Patterns

After you beat the two sesters, the Stray Sester will appear.

How to Beat the Twin Sesters and Stray Sesters
Twin Sesters
[1] Focus on one at a time.
The Red Sester acts fast and will keep attacking if you do not get rid of the Black Sester right away so focusing on the Red Sester first might be better if you don't have good damage.
But if you can easily get rid of the Black Sester, do so as she can do long-ranged attacks.
[2] Pile on the attacks.
Piling on the attacks and using Riposte to your advantage will make this an easy fight.
Stray Sesters
[3] Learn her patterns.
Create some distance when you see that she is about to strike since her multi-hit attacks can be deadly. Repel her attacks if you can with Perfect Guard/Harden/Parry.
